Tato diplomová práce se zabývá výběrem a optimalizací analytické metody vhodné pro stanovení vybraných biogenních aminů (putrescinu, histaminu, tyraminu a tryptaminu). V rámci optimalizace a validace metody byla prováděna měření, jak čistých standardů, tak standardů derivatizovaných dansylchloridem. Výsledkem těchto měření bylo zjištění, že derivatizovaná forma biogenních aminů je vhodnější pro stanovení metodou ultraúčinné kapalinové chromatografie (UPLC) ve spojení s hmotnostním spektrometrem (MS) jako detektorem. Následně byla metoda kapalinové chromatografie s tandemovou hmotnostně-spektrometrickou detekcí využita ke stanovení sledovaných biologicky aktivních látek ve vzorcích sýru, který byl podroben skladovacímu experimentu. Získané výsledky byly zpracovány a interpretovány v rámci této diplomové práce. |

The aim of this thesis was selection and optimization of suitable analytical method for analysis of biogenic amines (putrescine, histamine, tyramine and tryptamine). The optimization and validation of this method were made by measurements of pure standards and derivatized standards with dansyl chloride. The result of these measurements was that the derivatized form of biogenic amines is more suitable for the analysis by ultra high performance liquid chromatography method (UPLC) in connection with tandem mass spectrometry (MS/MS). Subsequently, the metod UPLC-MS/MS was used to determine monitored biologically active compound in sample of cheese which has been subjected to the storage experiment. The results were processed and interpreted in this thesis |
